Semax
Semax is a synthetic heptapeptide derived from a fragment of adrenocorticotropic hormone (ACTH 4-10), originally developed in Russia as a nootropic and neuroprotective research compound. It has been studied for cognitive support, attention, and recovery after ischemic brain injury, with decades of published research from Eastern Europe and more limited independent Western data. Semax lacks the hormonal, corticosteroid-stimulating activity of native ACTH while retaining central nervous system effects attributed largely to its proline-glycine-proline (PGP) fragment. It is classified as a research compound in most Western jurisdictions and is not approved by the FDA for any use. Available human evidence comes largely from small trials conducted outside the United States, so conclusions about efficacy and long-term safety remain preliminary and should not be treated as medical guidance.
Reported benefits
- Associated in research with improved attention and short-term memory performance
- Studied for potential neuroprotective effects following cerebral ischemia
- May support mood stability and reduced stress-related symptoms in some studies
- Explored for supporting cognitive recovery after brain injury
- Reported non-sedating profile compared with some classic nootropic agents
How it works
Semax is thought to act primarily by upregulating expression of brain-derived neurotrophic factor (BDNF) and modulating central dopaminergic and serotonergic signaling, which may support synaptic plasticity and attention-related circuits. Its PGP metabolite is proposed to carry much of the compound's neuroactive effect independent of the parent peptide. Some research also points to effects on cerebral blood flow and antioxidant activity in neural tissue. These mechanisms are drawn largely from animal and in vitro models, and their translation to humans is not fully established.

Read before using
This tool produces a rough, educational estimate by scaling commonly-published research ranges to lean body mass. It is not a prescription, medical advice, or a substitute for a licensed clinician. Most peptides use fixed dosing, not weight-based dosing. Individual needs vary. You are solely responsible for how you use this information.

What is Semax used for?
Semax is a synthetic heptapeptide derived from a fragment of adrenocorticotropic hormone (ACTH 4-10), originally developed in Russia as a nootropic and neuroprotective research compound. It has been studied for cognitive support, attention, and recovery after ischemic brain injury, with decades of published research from Eastern Europe and more limited independent Western data. It is classified as a research compound in most Western jurisdictions and is not approved by the FDA for any use. Available human evidence comes largely from small trials conducted outside the United States, so conclusions about efficacy and long-term safety remain preliminary and should not be treated as medical guidance. Reported benefits include associated in research with improved attention and short-term memory performance, studied for potential neuroprotective effects following cerebral ischemia, may support mood stability and reduced stress-related symptoms in some studies.
What is a typical Semax dose?
Reported ranges run 200-600 mcg. A low protocol is 200 mcg once daily, and a high protocol is 600 mcg split into two daily doses. This is reference information, not a dosing recommendation.
What is the half life of Semax?
Plasma half-life is very short, estimated at well under an hour due to rapid enzymatic degradation, though biologically active metabolites may prolong functional effects for several hours.. That is what sets the dosing frequency, and it is why the timing guidance is morning.
How is Semax administered?
Subcutaneous: into fat, under the skin. Historically administered intranasally in Russian clinical and research use; this profile reflects subcutaneous injection as used in current research contexts.
How long is a Semax cycle?
4-6 weeks on, followed by 2-4 weeks off. Cycling is commonly discussed in research and vendor literature to limit theoretical tolerance, but no controlled human studies have established an optimal on/off schedule.
What are the side effects of Semax?
Reported side effects include headache or irritability reported in some users, injection site irritation with subcutaneous administration, anxiety or overstimulation at higher doses in anecdotal reports, insufficient long-term human safety data to characterize rare risks. Evidence level for this compound is well studied.
