Pasireotide
Also known as Signifor
Pasireotide is a multi-receptor somatostatin analog approved for Cushing's disease and acromegaly, with a broader somatostatin receptor binding profile (SSTR1, 2, 3, and 5) than octreotide or lanreotide. Its stronger SSTR5 affinity makes it particularly relevant for suppressing ACTH secretion in Cushing's disease driven by pituitary corticotroph tumors. It is used both as first-line therapy in select patients and as an option after surgery or other somatostatin analogs have failed.
Reported benefits
- Reduces cortisol excess in Cushing's disease
- Normalizes GH and IGF-1 in acromegaly
- Option for patients unresponsive to other somatostatin analogs
- Available in both immediate-release and long-acting depot forms
How it works
Pasireotide binds multiple somatostatin receptor subtypes, especially SSTR5, suppressing ACTH release from pituitary corticotroph tumors in Cushing's disease and reducing GH/IGF-1 secretion in acromegaly, via Gi-coupled inhibition of hormone release.

Read before using
This tool produces a rough, educational estimate by scaling commonly-published research ranges to lean body mass. It is not a prescription, medical advice, or a substitute for a licensed clinician. Most peptides use fixed dosing, not weight-based dosing. Individual needs vary. You are solely responsible for how you use this information.

Common questions
What is Pasireotide used for?
Pasireotide is a multi-receptor somatostatin analog approved for Cushing's disease and acromegaly, with a broader somatostatin receptor binding profile (SSTR1, 2, 3, and 5) than octreotide or lanreotide. Its stronger SSTR5 affinity makes it particularly relevant for suppressing ACTH secretion in Cushing's disease driven by pituitary corticotroph tumors. It is used both as first-line therapy in select patients and as an option after surgery or other somatostatin analogs have failed. Reported benefits include reduces cortisol excess in cushing's disease, normalizes gh and igf-1 in acromegaly, option for patients unresponsive to other somatostatin analogs.
What is a typical Pasireotide dose?
Reported ranges run 300-900 mcg. A low protocol is 300 mcg twice daily, subcutaneous, and a high protocol is 900 mcg twice daily, maximum labeled subcutaneous dose. This is reference information, not a dosing recommendation.
What is the half life of Pasireotide?
Approximately 12 hours for the immediate-release subcutaneous formulation. That is what sets the dosing frequency, and it is why the timing guidance is twice daily (am & pm).
How is Pasireotide administered?
Subcutaneous: into fat, under the skin. Immediate-release pasireotide is given subcutaneously twice daily; a long-acting intramuscular depot formulation also exists.
How long is a Pasireotide cycle?
Continuous twice-daily dosing or monthly depot, typically long-term per condition on, followed by Discontinued per prescriber decision off.
What are the side effects of Pasireotide?
Reported side effects include hyperglycemia or new-onset diabetes, more pronounced than with other somatostatin analogs, gastrointestinal upset, gallstones, injection site reactions, qt interval prolongation risk. Evidence level for this compound is well studied.
