Leuprolide

Also known as Leuprorelin, Lupron, Eligard

Leuprolide is a long-acting synthetic GnRH agonist delivered as a depot injection, among the most widely used drugs in its class. It is approved for advanced prostate cancer, endometriosis, uterine fibroids, and central precocious puberty. Like other GnRH agonists, it causes an initial gonadotropin/testosterone or estrogen surge before sustained suppression develops over several weeks. Depot strengths allow dosing intervals from monthly to twice yearly.

Reported benefits

  • Androgen deprivation therapy in advanced/metastatic prostate cancer
  • Symptom control in endometriosis and uterine fibroids
  • Management of central precocious puberty
  • Used as an adjunct to radiation therapy in localized prostate cancer

How it works

Chronic exposure to leuprolide desensitizes pituitary GnRH receptors, reducing LH and FSH secretion and consequently lowering testosterone or estrogen to castrate or menopausal levels after an initial stimulatory flare phase.

Read before using

This tool produces a rough, educational estimate by scaling commonly-published research ranges to lean body mass. It is not a prescription, medical advice, or a substitute for a licensed clinician. Most peptides use fixed dosing, not weight-based dosing. Individual needs vary. You are solely responsible for how you use this information.

This dose needs 450 units, more than a 100-unit syringe holds. Add more water, split the dose, or use a larger syringe.

Common questions

What is Leuprolide used for?

Leuprolide is a long-acting synthetic GnRH agonist delivered as a depot injection, among the most widely used drugs in its class. It is approved for advanced prostate cancer, endometriosis, uterine fibroids, and central precocious puberty. Like other GnRH agonists, it causes an initial gonadotropin/testosterone or estrogen surge before sustained suppression develops over several weeks. Depot strengths allow dosing intervals from monthly to twice yearly. Reported benefits include androgen deprivation therapy in advanced/metastatic prostate cancer, symptom control in endometriosis and uterine fibroids, management of central precocious puberty.

What is a typical Leuprolide dose?

Reported ranges run 3.75-45 mg. A low protocol is 3.75 mg once monthly depot, and a high protocol is 45 mg once every 6 months depot. This is reference information, not a dosing recommendation.

What is the half life of Leuprolide?

Parent compound approximately 3 hours; depot formulations provide sustained release over 1 to 6 months. That is what sets the dosing frequency, and it is why the timing guidance is as directed by your clinician.

How is Leuprolide administered?

Intramuscular: into muscle. Available as intramuscular or subcutaneous depot formulations depending on brand (e.g., Lupron Depot is IM, Eligard is subcutaneous); dosing interval is set by depot strength.

How long is a Leuprolide cycle?

Continuous per oncology protocol; often long-term or indefinite for metastatic disease on, followed by Discontinued per prescriber decision off. Duration is individualized by the treating physician; not a self-cycled peptide.

What are the side effects of Leuprolide?

Reported side effects include initial symptom/tumor flare, hot flashes, bone mineral density loss with long-term use, injection site reactions, mood changes. Evidence level for this compound is well studied.
