IGF-1 DES
Also known as Des(1-3) IGF-1, DES IGF-1
IGF-1 DES is a truncated form of insulin-like growth factor 1 missing the first three amino acids of the N-terminus, a modification that markedly reduces its affinity for IGF-binding proteins. This gives it substantially greater potency at the IGF-1 receptor relative to native IGF-1, but only for a very short window, since it is cleared from circulation quickly. It is studied primarily for localized, site-specific muscle growth when injected directly into a trained muscle. Human clinical data are limited, and most supporting evidence comes from animal and in vitro studies.
Reported benefits
- High potency at the IGF-1 receptor per unit dose
- Proposed localized hypertrophic effect at the injection site
- Short duration of action may allow more precise timing around training sessions
How it works
By removing the binding-protein-interacting N-terminal residues, IGF-1 DES remains largely free and unbound in tissue, allowing it to activate the IGF-1 receptor and downstream PI3K/Akt/mTOR signaling with greater immediate potency than native IGF-1. Its rapid clearance means this activation is brief and localized, which is why it is typically injected directly at the target muscle rather than administered systemically.

Read before using
This tool produces a rough, educational estimate by scaling commonly-published research ranges to lean body mass. It is not a prescription, medical advice, or a substitute for a licensed clinician. Most peptides use fixed dosing, not weight-based dosing. Individual needs vary. You are solely responsible for how you use this information.

Common questions
What is IGF-1 DES used for?
IGF-1 DES is a truncated form of insulin-like growth factor 1 missing the first three amino acids of the N-terminus, a modification that markedly reduces its affinity for IGF-binding proteins. This gives it substantially greater potency at the IGF-1 receptor relative to native IGF-1, but only for a very short window, since it is cleared from circulation quickly. It is studied primarily for localized, site-specific muscle growth when injected directly into a trained muscle. Human clinical data are limited, and most supporting evidence comes from animal and in vitro studies. Reported benefits include high potency at the igf-1 receptor per unit dose, proposed localized hypertrophic effect at the injection site, short duration of action may allow more precise timing around training sessions.
What is a typical IGF-1 DES dose?
Reported ranges run 20-60 mcg. A low protocol is 20 mcg once daily on training days, and a high protocol is 60 mcg once daily on training days. This is reference information, not a dosing recommendation.
What is the half life of IGF-1 DES?
Approximately 20-30 minutes. That is what sets the dosing frequency, and it is why the timing guidance is ~20 min before a workout.
How is IGF-1 DES administered?
Intramuscular: into muscle. Typically injected directly into the trained muscle immediately after exercise because of its very short half-life; some protocols use subcutaneous injection instead.
How long is a IGF-1 DES cycle?
4-6 weeks on, followed by 4-6 weeks off. Because of its localized, acute action, cycling is typically framed around training blocks rather than continuous administration.
What are the side effects of IGF-1 DES?
Reported side effects include localized swelling or discomfort at the injection site, hypoglycemia risk, particularly if combined with insulin, theoretical concern for promoting abnormal cell growth given potent growth-factor activity, very short half-life limits the window for effect, which some view as a practical drawback. Evidence level for this compound is emerging research.
