FOXO4-DRI

FOXO4-DRI is a synthetic cell-penetrating peptide engineered to disrupt the interaction between FOXO4 and p53 inside senescent cells. It emerged from preclinical aging research published in the mid-2010s, most notably a widely cited mouse study associating it with reduced markers of cellular senescence. Human clinical data remain essentially absent, and FOXO4-DRI is a highly experimental research compound rather than an established therapeutic. Claims about senolytic effects in humans are extrapolated from limited animal and cell-culture work and should be treated with significant caution.

Reported benefits

  • Investigated in preclinical models for selective clearance of senescent cells
  • Studied as a potential approach to reduce senescence-associated inflammatory markers
  • Explored in early research on tissue and organ function in aged animal models

How it works

FOXO4-DRI is a D-retro-inverso peptide designed to mimic the FOXO4 binding domain, competitively blocking FOXO4 from sequestering p53 inside senescent cells. This disruption is proposed to free p53 to initiate apoptosis selectively in senescent cells while sparing healthy cells. Selective clearance of senescent cells (senolysis) is the central mechanism under investigation.

Read before using

This tool produces a rough, educational estimate by scaling commonly-published research ranges to lean body mass. It is not a prescription, medical advice, or a substitute for a licensed clinician. Most peptides use fixed dosing, not weight-based dosing. Individual needs vary. You are solely responsible for how you use this information.

Common questions

What is FOXO4-DRI used for?

FOXO4-DRI is a synthetic cell-penetrating peptide engineered to disrupt the interaction between FOXO4 and p53 inside senescent cells. It emerged from preclinical aging research published in the mid-2010s, most notably a widely cited mouse study associating it with reduced markers of cellular senescence. Human clinical data remain essentially absent, and FOXO4-DRI is a highly experimental research compound rather than an established therapeutic. Claims about senolytic effects in humans are extrapolated from limited animal and cell-culture work and should be treated with significant caution. Reported benefits include investigated in preclinical models for selective clearance of senescent cells, studied as a potential approach to reduce senescence-associated inflammatory markers, explored in early research on tissue and organ function in aged animal models.

What is a typical FOXO4-DRI dose?

Reported ranges run 2.5-10 mg. A low protocol is 2.5 mg daily for 3-5 days, and a high protocol is 10 mg daily for 5 days. This is reference information, not a dosing recommendation.

What is the half life of FOXO4-DRI?

Not well characterized in humans; animal data suggest a short circulating half-life of a few hours. That is what sets the dosing frequency, and it is why the timing guidance is morning.

How is FOXO4-DRI administered?

Subcutaneous: into fat, under the skin.

How long is a FOXO4-DRI cycle?

3-5 consecutive days on, followed by 8-12 weeks or longer off. Proposed pulsed dosing reflects senolytic hit-and-run strategies used in preclinical models, not validated human protocols

What are the side effects of FOXO4-DRI?

Reported side effects include human safety profile not established, theoretical risk of unintended apoptosis in non-senescent tissue, injection site reactions reported anecdotally, unknown long-term effects. Evidence level for this compound is emerging research.
