What TRIUMPH 1 found
Eli Lilly's TRIUMPH 1 trial, a Phase 3, placebo controlled study of retatrutide in 2,339 adults with obesity, reported strong topline results, according to PharmExec. Over 80 weeks, the highest dose group lost an average of 28.3% of body weight, with close to half losing 30% or more. This follows an earlier Phase 2 trial of retatrutide published in the New England Journal of Medicine, which first reported the compound's outsized weight loss signal in a smaller, controlled group.
Retatrutide is a triple hormone receptor agonist targeting GLP-1, GIP, and glucagon receptors at once. It remains investigational. Lilly has not filed for FDA approval, though a submission could come this year. Side effect discontinuation rose with dose, from 4.1% to 11.3%, versus 4.9% on placebo. Full trial details are listed on ClinicalTrials.gov as NCT05929066.
Common questions
Is retatrutide FDA approved? No. TRIUMPH 1 is a large Phase 3 human trial. Retatrutide is not a marketed medication.
How is it different from approved GLP-1 drugs? It adds a third target, the glucagon receptor, beyond the GLP-1 and GIP pathways used by approved drugs.
Did every trial participant lose the same weight? No. Weight loss and side effect rates both rose with dose.
