The FDA has granted accelerated approval to Rybrevant Faspro (amivantamab and hyaluronidase lpuj) for adults with recurrent or metastatic head and neck squamous cell carcinoma whose disease progressed after platinum based chemotherapy and immunotherapy, according to an FDA press announcement. This is the first new approval in this setting in a decade, and the drug is not indicated for HPV associated oropharyngeal cancer.

The decision rests on an open label trial of 102 patients that found a 42% overall response rate, more than double the roughly 20% or less seen with currently approved therapies. Among responders, 56% maintained that response for six months or longer. Because accelerated approval relies on surrogate measures like response rate rather than confirmed survival benefit, the FDA is requiring Janssen Biotech to run confirmatory studies to verify long term clinical benefit.

The prescribing information carries warnings for hypersensitivity and infusion related reactions, interstitial lung disease, skin reactions, eye toxicity, and risk to a developing fetus. The drug previously received Breakthrough Therapy designation for this use.

This is an approved prescription oncology drug, not a research peptide, and the news is included here as a notable development in clinical oncology rather than a product available for general use.
