Blocking an Aging Protein Regrew Cartilage in Mice

Researchers at Stanford Medicine report that blocking a protein called 15 PGDH restored worn cartilage in aging mice and helped protect their knees from arthritis after ACL like injuries, according to a Stanford Medicine summary on ScienceDaily. The protein is described as a gerozyme, an enzyme that becomes more abundant with age and is linked to loss of function in muscle, bone, nerve and blood tissue.

What stands out here is the mechanism. In other tissues, repair after blocking 15 PGDH depends on stem cells multiplying. In cartilage, the existing chondrocytes themselves shifted their gene activity toward a more youthful pattern, without new stem cell involvement. The team, led by Helen Blau and Nidhi Bhutani, also exposed human cartilage collected during knee replacement surgeries to the same inhibitor and saw it begin producing new, functional cartilage tissue.

This remains animal and laboratory tissue research, published in Science, not a tested human therapy. Osteoarthritis currently has no approved drug that reverses cartilage loss, only treatments that manage pain and swelling. The researchers suggest the approach could eventually lead to an oral medicine or injection, but that would require clinical trials in people to confirm safety and effectiveness before anything resembling medical use.
