# Gonadorelin

> Gonadorelin is a synthetic decapeptide identical in sequence to endogenous gonadotropin-releasing hormone (GnRH). It is used diagnostically to assess pituitary responsiveness and clinically to help maintain endogenous testicular function and fertility potential in men undergoing testosterone replacement or other gonadal-suppressive therapies. Because its action depends on pulsatile rather than continuous exposure, dosing frequency and pattern matter more than with many peptides. It is a prescription hormone product, not a general wellness supplement.

- Dose range: 100-500 mcg
- Dose tiers: low 100 mcg; moderate 250 mcg; high 500 mcg
- Half-life: Approximately 10 to 40 minutes
- Timing: As directed by your clinician
- Cycle: Ongoing alongside testosterone replacement or other suppressive therapy, per prescriber on, As directed if TRT or the suppressive agent is discontinued off
- Storage: Refrigerate lyophilized powder at 2-8°C; reconstituted solution should be refrigerated and used within the timeframe specified by the manufacturer.

## Mechanism of action

Gonadorelin binds GnRH receptors on anterior pituitary gonadotrope cells. Pulsatile exposure stimulates release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which in turn drive gonadal steroidogenesis and gametogenesis. Continuous, non-pulsatile exposure instead downregulates GnRH receptors and suppresses gonadotropin output, which is the basis for its diagnostic and, in other agents of this class, therapeutic-suppression uses.

## Reported benefits

- Assesses pituitary-gonadal axis function diagnostically
- Helps maintain testicular size and endogenous testosterone/sperm production during TRT
- Supports fertility-preservation protocols in hypogonadal men
- Used in pulsatile-pump therapy for hypogonadotropic hypogonadism

## Reported side effects

- Injection site reactions
- Headache
- Flushing
- Nausea
- Rare hypersensitivity reactions

## Common protocols

- GnRH stimulation test (diagnostic)
- Adjunct during testosterone replacement therapy to preserve fertility
- Pulsatile pump therapy for hypogonadotropic hypogonadism

## Stacking notes

Frequently combined with exogenous testosterone therapy specifically to preserve testicular volume and fertility potential.
