# FOXO4-DRI

> FOXO4-DRI is a synthetic cell-penetrating peptide engineered to disrupt the interaction between FOXO4 and p53 inside senescent cells. It emerged from preclinical aging research published in the mid-2010s, most notably a widely cited mouse study associating it with reduced markers of cellular senescence. Human clinical data remain essentially absent, and FOXO4-DRI is a highly experimental research compound rather than an established therapeutic. Claims about senolytic effects in humans are extrapolated from limited animal and cell-culture work and should be treated with significant caution.

- Dose range: 2.5-10 mg
- Dose tiers: low 2.5 mg; moderate 5 mg; high 10 mg
- Half-life: Not well characterized in humans; animal data suggest a short circulating half-life of a few hours
- Timing: Morning

- Storage: Store lyophilized peptide frozen; reconstituted solution refrigerated and used promptly

## Mechanism of action

FOXO4-DRI is a D-retro-inverso peptide designed to mimic the FOXO4 binding domain, competitively blocking FOXO4 from sequestering p53 inside senescent cells. This disruption is proposed to free p53 to initiate apoptosis selectively in senescent cells while sparing healthy cells. Selective clearance of senescent cells (senolysis) is the central mechanism under investigation.

## Reported benefits

- Investigated in preclinical models for selective clearance of senescent cells
- Studied as a potential approach to reduce senescence-associated inflammatory markers
- Explored in early research on tissue and organ function in aged animal models

## Reported side effects

- Human safety profile not established
- Theoretical risk of unintended apoptosis in non-senescent tissue
- Injection site reactions reported anecdotally
- Unknown long-term effects

## Common protocols

- Short pulsed courses of a few consecutive days, repeated infrequently, modeled on preclinical senolytic dosing strategies

## Stacking notes

Sometimes discussed alongside other senolytic or mitochondrial-support peptides such as SS-31; no human interaction data exists
