# Alpha-MSH

> Alpha-MSH is a naturally occurring 13-amino-acid peptide hormone derived from proopiomelanocortin, best known for stimulating melanin production but also studied for broad anti-inflammatory and immunomodulatory properties. Research spans its roles in skin pigmentation, appetite regulation, and systemic inflammation control. Synthetic analogs of alpha-MSH have been explored clinically for conditions such as erythropoietic protoporphyria, though alpha-MSH itself is used mainly in research settings. Effects can vary meaningfully depending on which melanocortin receptor subtype is engaged.

- Also known as: Alpha-Melanocyte-Stimulating Hormone
- Primary category: Anti-Inflammatory
- Effectiveness rank: #6 in Anti-Inflammatory
- Evidence level: Moderate evidence
- Route: Subcutaneous
- Dose range: 250-1,000 mcg
- Dose tiers: low 250 mcg; moderate 500 mcg; high 1000 mcg
- Half-life: Short, estimated at approximately 30-60 minutes
- Timing: Morning
- Cycle: 4-6 weeks on, 4 weeks off (Cycling is commonly used to limit pigmentation buildup and reassess tolerance)
- Storage: Store lyophilized peptide frozen; reconstituted solution refrigerated and used within 2-3 weeks

## Mechanism of action

Alpha-MSH binds melanocortin receptors, particularly MC1R and MC3R, which are expressed on immune cells as well as melanocytes. Engagement of these receptors is associated with downregulation of pro-inflammatory cytokines and suppression of NF-kB activity, contributing to its studied anti-inflammatory profile. Through MC1R on melanocytes, it also stimulates melanogenesis, which accounts for its tanning-related use.

## Reported benefits

- Studied for broad anti-inflammatory and immunomodulatory effects
- Explored for skin pigmentation and photoprotection research
- Investigated in models of systemic inflammatory conditions
- May support appetite and stress-response regulation via central melanocortin pathways

## Reported side effects

- Nausea, especially at higher doses
- Flushing
- Increased skin pigmentation (may be undesired outside intended use)
- Appetite changes
- Rare mood or libido effects related to central melanocortin activity

## Common protocols

- Daily or every-other-day subcutaneous dosing in short cycles

## Stacking notes

Sometimes discussed alongside melanotan analogs for pigmentation research, and alongside KPV for anti-inflammatory focus
